This functionality already in  Deeplink Module. Deeplink module has a one HitCount Attribute that one help to achieve this functionality. When user click the DeepLink Url the Hitcount will be  increasing Automatically. So you can write a Decision to Check whether this link already used or not by Using the HitCount Attribute. And you can write your logic if it has used.

Not sure, if i understood your question correctly.

You want to disable URL once it is visited- You can write a logic in microflow that if url visited, change some boolean value to true and check boolean next time if it is true. if yes then redirect to some other page which will show you information message .microflow should call from deeplink handeler microflow.
